Linkage maps of Eucalyptus globulus using RAPD and microsatellite markers

Abstract
We report here the construction of RAPD maps based on an F1 intraprovenance cross in Eucalyptus globulus subsp. globulus. In addition a number of microsatellite loci originating from E. globulus and four other Eucalyptus species, have been added to the RAPD maps allowing the determination of homology between most linkage groups of the two parents. Homology with several linkage groups from maps of E. grandis and E. urophylla has also been established based on the sharing of microsatellite loci. At a LOD score threshold of 4.9 the male parent has 13 linkage groups covering 1013cM with 101 framework markers ordered at LOD 3.0. The female parent has 11 linkage groups covering 701cM with 97 framework markers. Eighteen microsatellite loci have been placed on the male parental map with thirteen loci placed on the female map. On the female map in particular there were more regions of segregation distortion than expected and genetic mechanisms which have been proposed to explain distorted segregation are discussed.
